Nucleus Sagulum

After multiple injections of wheat germ agglutinin conjugated to horseradish peroxidase (WGA-HRP) into the SC, the heaviest concentrations of labelled cells were found in the nucleus of the brachium (BIN) and external nucleus of the inferior colliculus, with much weaker labelling in the nucleus sagulum, dorsal, intermediate and ventral nuclei of the lateral lemniscus, paralemniscal regions, and periolivary nuclei.

A low density was found in the superior and inferior colliculi, the interpeduncular nucleus, the nucleus sagulum, the superior central nucleus, the cuneiform nucleus, the accessory dorsal tegmental nucleus, the nucleus of the solitary tract, the dorsal motor nucleus of the vagus, and the paralemniscal, magnocellular, gigantocellular, and lateral tegmental fields.

Descending fibers from the caudal GP course in the cerebral peduncle and project to posterior thalamic nuclei (the subparafascicular and suprageniculate nuclei, medial division of the medial geniculate nucleus, and posterior intralaminar nucleus/peripeduncular area) and to extensive brainstem territories, including the pars lateralis of the substantia nigra, lateral terminal nucleus of the accessory optic system, nucleus of the brachium of the inferior colliculus, nucleus sagulum, external cortical nucleus of the inferior colliculus, cuneiform nucleus, and periaqueductal gray.

To a lesser extent it was found in the nucleus sagulum and in the area medial to the lemniscal nuclei.

Injections of D-[ 3H]aspartate suggest that glutamatergic endings in the IC arise primarily from intrinsic connections, the opposite IC, layer 5 of temporal cortex, nucleus sagulum, and lateral lemniscal nuclei.

In the lateral tegmental field, the marginal nucleus of the brachium conjunctivum, the superior central nucleus, the nucleus sagulum, the dorsal nucleus of the raphe, the interpeduncular nucleus and the retrorubral nucleus the density of immunoreactive cell bodies was moderate. A moderate density of calbindin-immunoreactive fibers was found in the retrorubral nucleus, the central linear nucleus, the locus coeruleus, the nucleus sagulum, the dorsal nucleus of the raphe, the cuneiform nucleus, the ventral and dorsal nuclei of the lateral lemniscus, the medial nucleus of the solitary tract, the dorsal motor nucleus of the vagus, and the cuneate nucleus.

The periaqueductal gray, brachium of the inferior colliculus, nucleus of the brachium of the inferior colliculus, locus coeruleus, nucleus incertus, Kölliker-Fuse nucleus, facial nucleus, medial nucleus of the solitary tract and the area postrema contained a moderate density of immunoreactive fibres, whereas the pericentral nucleus of the inferior colliculus, nucleus sagulum, cuneiform nucleus, dorsal nucleus of the raphe, superior central nucleus, central, lateral and paralemniscal tegmental fields, ventral nucleus of the lateral lemniscus, dorsal tegmental nucleus, postpyramidal nucleus of the raphe, nucleus ambiguus, accessory dorsal tegmental nucleus, dorsal motor nucleus of the vagus and the inferior olive had the lowest density of immunoreactive fibres..

By contrast, the interpeduncular nucleus, magnocellular part of the red nucleus, central tegmental field, cuneiform nucleus, dorsal tegmental nucleus, nucleus sagulum and the medial and inferior vestibular nuclei had the lowest density, whereas a moderate density of immunoreactive cell bodies was found in the superior colliculus, medial division of the dorsal nucleus of the raphe, nucleus incertus, locus coeruleus and in the Kölliker-Fuse area. A moderate density of immunoreactive fibers was found in the dorsal motor nucleus of the vagus and in the postpyramidal nucleus of the raphe and a low density in the cuneiform nucleus, Kölliker-Fuse area, nucleus sagulum, inferior and superior central nuclei, lateral reticular nucleus and in the lateral and magnocellular tegmental fields..

The external cortex of the IC projects to the nucleus sagulum (Sag), the RPO, the VNTB, and the DCN.

Moreover, a high or moderate density of parvalbumin immunoreactive processes was visualized in the nucleus ruber, substantia nigra, superior and inferior colliculi, periaqueductal gray, nucleus sagulum, cuneiform nucleus, Kölliker-Fuse nucleus, nucleus of the trapezoid body, vestibular nuclei, dorsal motor nucleus of the vagus, and in the lateral reticular nucleus.

Cells of origin of these projections were localized in the caudal 2/3 of the GP, and their major target sites included the peripeduncular region, nucleus of the brachium of the inferior colliculus, para-lateral lemniscal zone, nucleus sagulum, external and pericentral nuclei of the inferior colliculus, and cuneiform nucleus.

A moderate density of cell bodies containing the peptide was observed in the ventral nucleus of the lateral lemniscus, accessory dorsal tegmental nucleus, retrofacial nucleus and in the lateral reticular nucleus, whereas a low density of such perikarya was found in the interpeduncular nucleus, nucleus incertus, nucleus sagulum, gigantocellular tegmental field, nucleus of the trapezoid body, nucleus praepositus hypoglosii, lateral and magnocellular tegmental fields, nucleus of the solitary tract, nucleus ambiguous and in the nucleus intercalatus. Finally, few immunoreactive fibers were visualized in the interpeduncular nucleus, cuneiform nucleus, locus coeruleus, nucleus incertus, superior and inferior central nuclei, nucleus sagulum, ventral nucleus of the lateral lemniscus, nucleus praepositus hypoglosii, medial vestibular nucleus, Kölliker-Fuse area, nucleus ambiguous, retrofacial nucleus, postpyramidal nucleus of the raphe, nucleus of the solitary tract, dorsal motor nucleus of the vagus, lateral reticular nucleus and laminar and alaminar spinal trigeminal nuclei..

The nucleus sagulum, an area of the midbrain tegmentum, has been considered a component of a lateral tegmental system within the ascending auditory pathway to the thalamus. In this study, connections of the nucleus sagulum within the midbrain were investigated in adult cats. The nucleus sagulum was identified as a region of principally small neurons (261 +/- 79 micron2) at the margin of the midbrain and neighboring the nuclei of the lateral lemniscus. Injections of tritiated leucine in the nucleus sagulum labeled axons that ended in dense patches within the superficial layers of the caudal portion of the dorsal cortex of the inferior colliculus on the ipsilateral side. Outside of the inferior colliculus, sagulum injections labeled other axons ending in the ventral intercollicular tegmentum on both sides and in a dorsal and rostral region of the contralateral nucleus sagulum that appeared contiguous with the dorsal nucleus of the lateral lemniscus. The distribution of axonal labeling after injections in the nucleus sagulum was contrasted with the distribution of projections from several neighboring areas of the lateral tegmentum, including the dorsal nucleus of the lateral lemniscus. None of these areas exhibited connections with the superficial layers of the caudal cortex of the inferior colliculus, which was the major target in the inferior colliculus of the nucleus sagulum. Thus, the results indicated that the nucleus sagulum is distinguished from adjacent regions of the lateral tegmentum by its connectivity.

The major inputs to the caudodorsal nucleus (DC) stem from nucleus sagulum and the pericentral nucleus of the inferior colliculus (ICP).

Midbrain degeneration was found bilaterally in IC, SC, nucleus sagulum, and tegmentum; a minor projection was noted in interpeduncular nucleus.

First, potential sources of auditory afferents were limited mainly to the external nucleus of the inferior colliculus, the nucleus sagulum, and the dorsomedial periolivary cell group.
